TRiPSoma and TRiPGermline
Ongoing research and the evolution of the Transgenic RNAi Project has led to two stock collections: TRiPSoma and TRiPGermline. You can find these stocks in one list here. The downloadable excel file indicates whether you can expect the hairpin to be expressed in the soma, germline, or both.
The TRiPSoma collection includes first generation stocks that were made in the VALIUM1 and VALIUM10 vectors using our long double-stranded hairpin approach, as well the second generation stocks constructed in the VALIUM20 vector using short hairpin microRNA technology. These lines have proven effective for transcript knockdown in somatic cells.
The TRiPGermline collection contains stocks made in the VALIUM20 and VALIUM22 vectors (also VALIUM21 - a variant of VALIUM22). VALIUM20 is great for knockdown in the soma and also works in the germline. VALIUM22 (and VALIUM21) has shown to be an exceptional vector for germline expression of RNAi.

Community Access to the TRiP Stocks
Our goal is for all TRiP Stocks to be available at the Bloomington Drosophila Stock Center (BDSC) at a reasonable time-point following production. If a TRiP stock has not yet been deposited at the BDSC, please contact Laura Holderbaum (lholderbaum@genetics.med.harvard.edu) and the TRiP facility at HMS will send it to you. If you are requesting more than five TRiP stocks please list them in an Excel file with a column for the CG# and a column for the TRiP#.
The TRiP welcomes feedback on the lines you have received. In particular, we appreciate information relevant to the efficiency and specificity of knockdown (e.g., whether or not an expected phenotype is observed). This information will allow us to troubleshoot and plan for future RNAi lines. Please send feedback to TRiP@flyrnai.org.
If use of any of the TRiP stocks results in data that is included in a manuscript for publication, the TRiP requests that a version of the following statement be included in the Acknowledgements section: "We thank the TRiP at Harvard Medical School (NIH/NIGMS R01-GM084947) for providing transgenic RNAi fly stocks used in this study."
What genes are already in production and how can I nominate
a gene(s)?
Please check the current list of TRiP stocks. If your gene of interest is not listed you may nominate it.
Genes will be added to the TRiP queue from three possible
avenues. (A) Selections will be in keeping with the BDSC
mandate of one mutation per gene. (B) We will meet the needs of
screeners at the Drosophila RNAi Screening Center (DRSC) for in
vivo follow-up studies subsequent to cell-based screens. And
(C) we will meet the needs of the Drosophila community for in
vivo phenotypic analyses through a nomination process. In
keeping with our successful proposal to NIH/NIGMS, priority
will go to nominations that help to fill in the phenotype gap
and overcome issues associated with pleiotropy.
To nominate a gene(s), the PI of the lab should send a
letter to TRiP@flyrnai.org that
includes an explanation of why the specific gene is being
nominated.
